does the neck weight on a chambered body affect guitar balance, standing OR sitting ?

In general yes it has an effect.  There are many other things that affect the hang angle as well, including weight of the tuners, location of strap button, neck profile, etc.  Weather or not all this creates something desirable or undesirable is going to be up to the specifics of your particular design.

Add to this the actual weight of the body, body shape, strap button locations, etc... and weight of pickups and hardware.  Those will act as a counterbalance.  You may or may not have any issues with neck dive.  Too broad of a question to give any more specifics.

so.... i would really need to know chambered body weight & neck weight w/tuners to see if it would balance ok ?

Basically it's like this:

My Jaguar body weighs 4.1 pounds. Add on all the hardware and it goes up, let's just say a pound for measuring reasons. That's 5.1 pounds. Proper location of strat buttons is important here.

Then we put the neck on, with the Vintage tuners. It's going to balance well because the neck does not outweigh the body.

However, if you take an SG body and add a neck with very heavy wood, say Wenge/Wenge, it will neck dive.

Please be aware I cited my own unfinished guitar body and neck as points of reference.

I don't know if this helps, but I have a three pound chambered Strat body with a Mahogany/Rosewood neck (not sure of weight).  When I bought it, I was concerned about the balance.  I remember my initial impression was that it may be slightly neck heavy; however, at this point I simply don't notice it at all.  For me, any minor inconvenience is way overshadowed by taking a pound or more off my shoulder (or knee), but of course, someone else might feel totally different about it.

For what it's worth, I compared the balance of my Warmoth Strat to my Tom Anderson.  They are basically the same. Either one will have the neck drop down when they are perched on my knee. So perhaps both are a little heavy on the neck end, but I've had my Anderson for over ten years and never noticed. So in my opinion, unless you're getting a really heavy neck compared to the body, you will probably not notice. Again only my opinion which isn't worth much.

Careful - the strat has the strap button located about three inches further down the neck than say an SG would.  This would make the strat OK and the SG dive into the ground.  Still need to take strap button location into consideration.

Dalo - it would really help if you told us what you were considering building.

looking at the strat chambered body, walnut body & neck w/pau ferro board. 2 hums, gotoh tuners. being chambered
and not semi hollow it might balance well. also thought about solid body w/swimming pool top rout. just trying to lighten up the walnut heft. THANKS for all the replies !
