I have a set of 6 Kluson Deluxe tuners in a bag with screws, bushings and washers. On my scale, they weigh 6.3 ounces, or 178 grams. I also have a set of 6 Schaller Mini-Lockers in a bag with screws, bushings and washers. They weigh 8.7 ounces, or 246 grams. So, replacing the cheapest, lightest, junkiest tuners extant with the most expensive, heaviest, best lockers would add 2.4 ounces to any guitar. Negligible, to say the least. Wearing a sweatshirt instead of a t-shirt would put more load on you.

You must have left a socket wrench or some locking pliers in your control cavity or something <grin>

Yeah, that's some funny stuff right there.

  I don't own a precision scale (or any scale at all, now that I think of it) to scientifically measure the mass of the string segments.

You also need to consider the weight savings with the lockers since you no longer have the additional winds of the strings on the post.
 
The weight isn't that noticeable, but the sustain is from the added mass.

My ole ryth gtr player had a 7 string Carvin with the Floyd setup, along with the Sperzels, and it sings for days.

I suspect your buddy's playing style had something to do with that, or the guitar itself had something else going on. Adding 2 ounces to the headstock isn't going to affect sustain to a noticeable degree.

 
I imagine its the much more direct coupling of the string to the tuner. Not only will there be transfer from the post but not the locking pin as well.  :dontknow:
 
I don't know how much credit I'd give the locking pin, but eliminating all those winds on the peg probably counts for something. It's essentially a spring. That's why "standard" tuners are so awful. So, how much? Good question. I suspect not much, but it hasn't been measured. Physics says it makes a difference, but that can be imperceptible to mere humans.

That's the problem with most guitar dogma - no reliable, objective measurement scheme. It's all based on hearsay, perception and theory. You would think the major manufacturers with jillions of dollars to play with might spend a few grand to do some definitive studies, but they don't (or don't share the data if they do). They behave like the pharmaceuticals: if you cure it, you wreck the market. So, don't cure it, treat it. If you just treat it, you've got a consistent revenue stream.
